FOXO4-DRI
FOXO4-DRI is a synthetic D-retro-inverso peptide that mimics the region of the transcription factor FOXO4 that interacts with the p53 protein. It is made of mirror-image D-form amino acids arranged in reverse order, which makes it more resistant to proteases than the natural L-analogue. PeptXBio supplies it as a lyophilised powder, ready for laboratory research.
Quality and purity
≥99% purity (HPLC). High-grade laboratory lyophilised powder with analytically verified purity and consistent batch quality, so that reconstitution and research results remain reproducible.

Why it is studied
FOXO4-DRI is used as a tool to study the FOXO4–p53 protein interaction in senescent cells in vitro. The peptide competes with FOXO4 for the p53 transactivation domain: NMR structural models show that both the FOXO4-derived part and the cationic cell-penetrating chain contribute to binding to the disordered p53TAD2. In this way, questions of senescence, senolytic mechanisms and the p53 signalling pathway are studied in cell cultures. This information is provided for research context only and is not an instruction for use.
Reconstitution
For laboratory work, the powder is usually dissolved in bacteriostatic water by gently swirling the vial until the solution is clear – do not shake, so as not to damage the peptide. The prepared stock solution is kept refrigerated and aliquoted according to the research protocol. The volume of solvent is chosen according to the desired working concentration in the experiment.

Frequently asked questions
FOXO4-DRI is a synthetic D-retro-inverso peptide that mimics the region of the transcription factor FOXO4 that interacts with the p53 protein. It consists of 46 mirror-image D-form amino acids arranged in reverse order, which makes it more resistant to proteases than the natural L-analogue. In the research context, the peptide acts as a molecular tool that competes with FOXO4 for binding to the p53 transactivation domain: by disrupting the FOXO4–p53 interaction, it allows the p53 signalling pathway in senescent cells to be studied in vitro. This information is provided for scientific context only and is not an instruction for use.
In cell cultures, FOXO4-DRI is used to study cellular senescence, senolytic mechanisms and the FOXO4–p53 protein interaction in senescent cells. NMR structural models show that both the FOXO4-derived part of the peptide and the cationic cell-penetrating chain contribute to binding to the disordered p53TAD2. The compound has been widely described as a tool in in vitro models of senolytic activity (Baar et al., Cell, 2017). The data are provided for laboratory research purposes only.
The product is supplied as a high-grade laboratory lyophilised (freeze-dried) powder of ≥99% purity (HPLC). Each vial contains 10 mg of FOXO4-DRI peptide. For laboratory work, the powder is usually reconstituted in bacteriostatic water, which is sold separately. Swirl the vial gently until the solution is clear, without shaking, so that the peptide is not damaged.
Keep the unopened vial of dry powder in a refrigerator (+2 to +8 °C), or in a freezer (−20 °C) for longer storage, protected from light. Once reconstituted in bacteriostatic water, keep the solution refrigerated and use it within the time set out in your research protocol. Proper storage helps preserve the stability of the peptide and the reproducibility of results.
In in vitro research models, FOXO4-DRI is most often studied in cultures of senescent cells – for example, human chondrocytes, fibroblasts or endothelial cells cultured in vitro in which senescence has been induced by damage or chemotherapeutic agents. Analytically, it is often compared against markers of the p53 signalling pathway and FOXO4 expression to assess senolytic activity in cell culture. The specific combination and concentrations are chosen according to the research protocol; no dosing recommendations for humans or animals are given.
Absolutely not. This product is sold exclusively for laboratory research and scientific experiments. It is not intended for human or animal consumption and is not a medicine, dietary supplement or cosmetic product.
