5-Amino-1MQ
5-Amino-1MQ (5-amino-1-methylquinolinium) is a synthetic, low-molecular-weight, membrane-permeable molecule of the methylquinolinium class. It is a selective inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT) and is widely used as a tool compound in biochemical research. It is not a peptide but a small organic molecule, supplied by PeptXBio as a lyophilised powder for laboratory work.
Quality and purity
≥99% purity (HPLC). Supplied as a high-grade laboratory lyophilised powder with analytically confirmed purity and uniform batch composition, so that reconstitution and research reproducibility are consistent.
Specifications
- CAS number: 42464-96-0
- Molecular formula: C₁₀H₁₁N₂ (cation)
- Molecular weight: 159.21 g/mol (cation)
- Chemical class: methylquinolinium small molecule (NNMT inhibitor)
Why it is studied
The NNMT enzyme uses S-adenosylmethionine (SAM) as a methyl donor to methylate nicotinamide, producing 1-methylnicotinamide and S-adenosylhomocysteine. The quinolinium scaffold of 5-Amino-1MQ is designed to bind the active site of NNMT while leaving other SAM-dependent enzyme systems largely unaffected. In in vitro adipocyte (fat cell) models, the effect of this inhibition on intracellular 1-methylnicotinamide levels, measured by LC/MS/MS, and on the cell’s pool of nicotinamide and methyl donors is studied. This is a research context of enzyme kinetics and metabolic pathways in cell and enzyme models, not an effect on the body. This information is provided for research context only and is not an instruction for use.
Reconstitution
For laboratory work, the powder is dissolved in bacteriostatic water (sold separately), letting the solvent run slowly down the side of the vial and swirling gently rather than shaking. The concentration of the dissolved stock solution is calculated from the amount in the vial and the volume of solvent added. The prepared solution is kept refrigerated and aliquoted only according to the research protocol under in vitro conditions.

Frequently asked questions
5-Amino-1MQ (5-amino-1-methylquinolinium) is a synthetic, low-molecular-weight, membrane-permeable molecule of the methylquinolinium class – not a peptide but a small organic molecule. In scientific research it is used as a selective inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). NNMT methylates nicotinamide using S-adenosylmethionine (SAM) as a methyl donor; the quinolinium scaffold of 5-Amino-1MQ binds the active site of NNMT while leaving other SAM-dependent enzyme systems largely unaffected. This makes it a tool compound for studying the mechanism of NNMT inhibition in vitro.
5-Amino-1MQ is used in research on enzyme kinetics and metabolic pathways in cell and enzyme models. In in vitro adipocyte (fat cell) models, the effect of NNMT inhibition on intracellular 1-methylnicotinamide levels, measured by LC/MS/MS, and on the cell’s pool of nicotinamide and methyl donors is studied. This in vitro context was described by Neelakantan and colleagues (Biochemical Pharmacology, 2018), where methylquinolinium NNMT inhibitors were membrane-permeable, relatively selective and reduced intracellular 1-methylnicotinamide in differentiated adipocyte cultures. This is a research context of enzyme and cell models, not an effect on the body.
The product is supplied as a high-grade laboratory lyophilised (freeze-dried) powder of ≥99% purity (HPLC). Each vial contains 50 mg of the active substance. For laboratory work, the powder is dissolved in bacteriostatic water, which is sold separately: the solvent is run slowly down the side of the vial and swirled gently rather than shaken. The concentration of the stock solution is calculated from the 50 mg in the vial and the volume of solvent added.
Keep the unopened vial of dry powder in a refrigerator (2–8 °C), or in a freezer (−20 °C) for longer storage, always protected from light. Once reconstituted in bacteriostatic water, keep the solution refrigerated and use it within the time set out in your research protocol. Avoid repeated warming and cooling cycles to keep research reproducibility consistent.
Because the action of 5-Amino-1MQ is linked to the NNMT / SAM / NAD+ metabolic pathway, in in vitro research it is most often analysed in models of differentiated adipocytes (e.g. cell lines) together with measurement of intracellular metabolites – 1-methylnicotinamide, NAD+ and SAM – by LC/MS/MS. In a methodological context, it is compared with the reference compound 1-methylquinolinium and with substrates of other SAM-dependent methyltransferases to assess the selectivity of inhibition. Specific concentrations and conditions are set only according to the in vitro research protocol; no doses for humans or animals are given.
Absolutely not. This product is sold exclusively for laboratory research and scientific experiments. It is not intended for human or animal consumption and is not a medicine, dietary supplement or cosmetic product.
